Spermatozoa Characteristics ef Streptozotocin-induced Diabetic Zucker Lean Rat:Calcium lonophore-induced Acrosome Reaction and Sperm Concentration



Abstract
This study was designed to investigate the changes of sperm concentration and the characteristics of acrosome reaction at epididymis and vas deference of diabetic zucker lean rat induced with streptozotocin(70mg/kg, ip). Experimental animal was sacrificed at 3 days and 14 days after the streptozotocin injection. Diabetic was induced in rats and the levels of insulin and glucose in serum showed a pattern of inverse proportion. ¢¥1¢¥he sperm concentration in epididymal caput was slowly decreased whole diabetic condition (control, 3 day and 14 day : 15.3+2.6, 8.2+0.7 and 7.6+0.8, respectively). The sperm concentration in epididymal corpus was significantly decreased at 3 days and 14 days, but that in epididymal cauda the concentration was significantly decreased at 14 days only. Spontaneous reaction was showed significantly difference between caudal and was deferencial sperm (respectively 38.1+1.2, 51.1+7.6). In diabetic groups, the spontaneous reaction rate was significantly increased in caudal sperm versus the control group. Spontaneous reaction rate of was deferential sperm was significantly increased at 14 days group. The ARIC (acrosome reaction to ionophore challinge) value of caudal sperm was range between 11.0 and 12.7% in all groups. From those results, we knew that the concentration of epididymal sperm was decreased in diabetic condition (appeared at first corpus and lately cauda and vas dererence) and the spontaneous reaction rate was increased at diabetic rat. It is suggested that diabetes mellitus induce the decrease in sperm concentration and inhibit the epididymal sperm maturation. It is thought that diabete mellitus may be associated with decreasement of fertilization rate and subsequent infertility.
